Prohibitin 1 is essential to preserve mitochondria and myelin integrity in Schwann cells

چکیده

Abstract In peripheral nerves, Schwann cells form myelin and provide trophic support to axons. We previously showed that the mitochondrial protein prohibitin 2 can localize axon-Schwann-cell interface is required for developmental myelination. Whether homologous 1 has a similar role, whether prohibitins also play important roles in cell mitochondria unknown. Here, we show deletion of minimally perturbs development, but later triggers severe demyelinating neuropathy. Moreover, are heavily affected by ablation demyelination occurs preferentially with apparent loss. Furthermore, response damage, trigger integrated stress response, but, contrary what was suggested, this not detrimental context. These results identify role integrity advance our understanding about damage.

S100 is preferentially distributed in myelin-forming Schwann cells.

In order to elucidate the role of S100 protein in vivo we used postembedding electron microscopical immunocytochemistry with commercially available antibodies to S100, and secondary antibodies conjugated to colloidal gold to determine the distribution of S100 protein in the sciatic nerve and the cervical sympathetic trunk of the rat. We found that S100 immunoreactivity in Schwann cells was diff...

Improved pre-embedded immuno-electron microscopy procedures to preserve myelin integrity in mammalian central nervous tissue

Preparation of specimens for pre-embedded immuno-electron microscopic analysis of CNS tissue is challenging. Conventional methods frequently result in significant damage to myelin integrity, ultimately confounding accurate analysis of myelin quality and assessment of changes in the number of lamellae. We have developed and optimized a protocol for maintaining good myelin morphology in CNS tissu...

mTORC1 promotes proliferation of immature Schwann cells and myelin growth of differentiated Schwann cells.

The myelination of axons in peripheral nerves requires precisely coordinated proliferation and differentiation of Schwann cells (SCs). We found that the activity of the mechanistic target of rapamycin complex 1 (mTORC1), a key signaling hub for the regulation of cellular growth and proliferation, is progressively extinguished as SCs differentiate during nerve development. To study the effects o...

The molecular machinery of myelin gene transcription in Schwann cells.

During late fetal life, Schwann cells in the peripheral nerves singled out by the larger axons will transit through a promyelinating stage before exiting the cell cycle and initiating myelin formation. A network of extra- and intracellular signaling pathways, regulating a transcriptional program of cell differentiation, governs this progression of cellular changes, culminating in a highly diffe...

Developmental changes in myelin-induced proliferation of cultured Schwann cells

Schwann cell proliferation induced by a myelin-enriched fraction was examined in vitro. Although nearly all the Schwann cells contained material that was recognized by antisera to myelin basic protein after 24 h, only 1% of the cells were synthesizing DNA. 72 h after the addition of the mitogen a maximum of 10% of the cells incorporated [3H]thymidine. If the cultures were treated with the myeli...
